Rett syndrome (RTT), a neurodevelopmental X-linked disorder, is the leading cause of mental retardation in females affecting 1/10,000 newborn girls. The laboratory of prof. Enrico Tongiorgi has undertaken a multi-year project to repurpose FDA-approved drugs in RTT. The lab has implemented a cellular model of Rett syndrome that replicates the dendritic atrophy typical of the disease (https://doi.org/10.3389/fncel.2014.00018, https://doi.org/10.1038/s41598-020-59268-w) and has isolated drugs that can restore the normal morphology of the RTT neurons. The fellow will define the effects and the drug mechanism of action on neural cells carrying different MeCP2 mutations, using transgenic mice or iPSCs-derived neurons from RTT patients and unaffected individuals. Cellular neurobiology experience is needed, and previous experience with NCS and/or iPSC-derived neurons preferred.
The laboratory offers training in bioinformatics, molecular biology, pharmacology and extensive expertise in microscopy including cellular morphology analysis, spectral confocal microscopy and access to recently acquired microscopes including Elyra7 associated with ST-EM for Correlative Microscopy and live imaging, and a brand-new 2Photon microscope of the core facility of the University of Trieste. The lab participates to the International Master Degree in Neuroscience which attracts highly skilled and motivated students and participates in the PhD in Molecular Biomedicine.
